چکیده

Methods Subjects underwent Baseline Challenge in an Environmental Exposure Chamber (EEC) on 4 consecutive days, consisting of 3-hour allergen exposures (Fel d1 50.19±3.70 ng/m3). Subjects scored four nasal and four ocular symptoms every 30 minutes each on a scale of 0-3. 202 subjects were randomised to placebo, 8x3nmol Cat Peptide Antigen Desensitisation (Cat-PAD) 2weeks(wk) apart or 4x6nmol Cat-PAD 4wk apart. Subjects re-attended the EEC for 4 consecutive days, of 3-hours, 18-22wk after the start of treatment. 50-54wk after commencement of treatment subjects were invited to participate in a blinded follow-on study without further dosing. 89 subjects were enrolled and had a further 4 consecutive days, of 3-hours in the EEC.

Immunotherapy – 2067. FEL d 1 peptide antigen desensitization safety and efficacy in a double-blind, placebo-controlled environmental exposure chamber study

Background Allergic rhinoconjunctivitis is an increasing problem worldwide with significant impact on quality of life and productivity. Sensitivity to cats accounts for 10-15% of the disease burden. Previous immunotherapy studies with two 27aa peptides were unsuccessful as a result of early and late phase responses. Cat Peptide Antigen Desensitisation (Cat-PAD) is a mixture of seven T-cell epit...

The major cat allergen, Fel d 1, in diagnosis and therapy.

Sensitization to cat is a common cause of allergic disease all over the world. Symptoms range from mild rhinoconjunctivitis to potentially life-threatening asthmatic exacerbations. In vivo and in vitro diagnostics of cat allergy is currently based on cat dander extract. As allergen extracts from natural sources are heterogeneous in composition, the allergen content may vary. With the introducti...

Displaying Fel d1 on virus-like particles prevents reactogenicity despite greatly enhanced immunogenicity: a novel therapy for cat allergy

Allergen-specific desensitization is the only disease-modifying therapy currently available for the treatment of allergies. These therapies require application of allergen over several years and some may induce life-threatening anaphylactic reactions. An ideal vaccine for desensitization should be highly immunogenic and should alleviate allergic symptoms upon few injections while being nonreact...

In silico identification of epitopes from house cat and dog proteins as peptide immunotherapy candidates based on human leukocyte antigen binding affinity

The objective of this descriptive study was to determine Felis domesticus (cat) and Canis familiaris (dog) protein epitopes that bind strongly to selected HLA class II alleles to identify synthetic vaccine candidate epitopes and to identify individuals/populations who are likely to respond to vaccines. FASTA amino acid sequences of experimentally validated allergenic proteins of house cat and d...

Two year persistent treatment effect in reducing nasal symptoms of cat allergy after 4 doses of Cat-PAD, the first in a new class of synthetic peptide immuno-regulatory epitopes

Background Treatment with Cat-PAD (also known as ToleroMune Cat, the first in a new class of synthetic peptide immuno-regulatory epitopes), in an Environmental Exposure Chamber (EEC) model of cat allergy showed a persistent treatment effect one year [1] and two years [2] after administration of only 4 injections over 12 weeks. Here we report the differences in Total Nasal Symptom Scores (TNSS) ...
